RESUMO

We have functionally evaluated a transcription factor CaMYB59 for its role in pepper immune responses to Ralstonia solanacearum attack and high temperature-high humidity (HTHH). Exposure to R. solanacearum inoculation (RSI) and HTHH resulted in up-regulation of this nucleus-localized TF. Function of this TF was confirmed by performing loss of function assay of CaMYB59 by VIGS (virus-induced gene silencing). Plants with silenced CaMYB59 displayed not only compromised pepper immunity against RSI but also impaired tolerance to HTHH along with decreased hypersensitive response (HR). This impairment in defense function was fully linked with low induction of stress-linked genes like CaPO2, CaPR1, CaAcc and thermo-tolerance linked CaHSP24 as well as CaHsfB2a. Conversely, transient overexpression of CaMYB59 enhanced pepper immunity. This reveals that CaMYB59 positively regulated host defense against RSI and HTHH by means of HR like mimic cell death, H2O2 production and up-regulation of defense as well as thermo-tolerance associated genes. These changes in attributes collectively confirm the role of CaMYB59 as a positive regulator of pepper immunity against R. solanacearum. We recommend that such positive regulation of pepper defense is dynamically supported by phyto-hormone signaling and transcriptional web of defense genes. These integrated and interlinked events stabilize plant growth and survival under abiotic and biotic stresses.

This study aimed to access the impact of soil polluted with petroleum (5, 10 g petroleum kg-1 soil) on Bermuda grass (Cynodon dactylon L.) with and without applied bacterial inoculants (Arthrobacter oxydans ITRH49 and Pseudomonas sp. MixRI75). Both soil and seed were given bacterial inoculation. The evaluated morphological parameters of Bermuda grass were fresh and dry weight. The results demonstrated that applied bacterial inoculants enhanced 5.4%, 20%, 28% and 6.4%, 21%, and 29% shoot and root fresh/dry weights in Bermuda grass under controlled environment. The biochemical analysis of shoot and root was affected deleteriously by the 10 g petroleum kg-1 soil pollution. Microbial inoculants enhanced the activities of enzymatic (catalase, peroxidase, glutathione reductase, ascorbate peroxidase, superoxide dismutase) and non-enzymatic (ɑ-tocopherols, proline, reduced glutathione, ascorbic acid) antioxidant to mitigate the toxic effects of ROS (H2O2) under hydrocarbon stressed condition. The maximum hydrocarbon degradation (75%) was recorded by Bermuda grass at 5 g petroleum kg-1 soil contamination. Moreover, bacterial persistence and alkane hydroxylase gene (alkB) abundance and expression were observed more in the root interior than in the rhizosphere and shoot interior of Bermuda grass. Subsequently, the microbe used a biological tool to propose that the application of plant growth-promoting bacteria would be the most favorable choice in petroleum hydrocarbon polluted soil to conquer the abiotic stress in plants and the effective removal of polyaromatic hydrocarbons in polluted soil.

The NRAMPs (natural resistance-associated macrophage proteins) are major transporters for the absorption and transport of metals like Pb, Zn, Mn, Fe, and Cd in plants. While NRAMP gene family members have been extensively studied as metal transporters in model and other plants, little information has been reported on their role in Triticum aestivum, particularly in response to Cd stress. Current study reported 13 NRAMP candidates in the genome of T. aestivum. Phylogenetic analysis divided these into three clades. Motif and gene structure study showed that members in the same clades shared the same location and pattern, which further supported the phylogenetic analysis. The analysis of cis-acting elements in promoter sequences of NRAMP genes in wheat identified stress-responsive transcription factor binding sites. Multiple sequence alignment identified the conservation of important residues. Based on RNA-seq and qRT-PCR analysis, Cd stress-responsive variations of TaNRAMP gene expression were reported. This study provides comprehensive data to understand the TaNRAMP gene family, its features, and its expression, which will be a helpful framework for functional research.

In the modern world, plastic trash has been recognized as a global issue, and studies on microplastics (MPs) in the marine and inland environments have previously been conducted. Marine ecosystems act as a bio-diverse ecosystem where coral reefs contribute to make a sound living of the coastal people by gathering natural resources. The current study indicates that MPs and heavy metals (HMs) accumulation to biofilm and organic matter through sedimentation, precipitation, adsorption, and desorption that may have potential effect on growth and development of coral reefs in the marine ecosystems. However, the knowledge of distribution, impact, mechanism, degradation, and association mechanisms between MPs and HMs in the natural environment may open a new window for conducting analytical research from an ecological viewpoint. The current study thus summarizes the types of marine samples with the analytical techniques, polymers of MPs, and their impact on corals and other marine biota. This study also identifies existing knowledge gaps and recommends fresh lines of inquiry in light of recent developments in MPs and HMs research on the marine ecosystems. Overall, the present study suggests a sustainable intervention for reducing MPs and HMs from the marine ecosystems by demonstrating their existence in water, sediment, fish, corals, and other biota, and their impending ecotoxicological impacts on the environment and human health. The impacts of MPs and HMs on coral reefs are critically assessed in this study in light of the most recent scientific knowledge, existing laws, and new suggestions to minimize their contamination in the marine ecosystems.

This study investigated the physiological and molecular responses of rice genotype '9311' to Cd stress and the mitigating effects of silicon oxide nanoparticles (SiO NPs). Cd exposure severely hindered plant growth, chlorophyll content, photosynthesis, and Cd accumulation. However, SiO NPs supplementation, particularly the SiONP100 treatment, significantly alleviated Cd-induced toxicity, mitigating the adverse effects on plant growth while maintaining chlorophyll content and photosynthetic attributes. The SiONP100 treatment also reduced Cd accumulation, indicating a preference for Si uptake in genotype 9311. Complex interactions among Cd, Si, Mg, Ca, and K were uncovered, with fluctuations in MDA and H2O2 contents. Distinct morphological changes in stomatal aperture and mesophyll cell structures were observed, including changes in starch granules, grana thylakoids, and osmophilic plastoglobuli. Moreover, following SiONP100 supplementation, genotype 9311 increased peroxidase, superoxide dismutase, and catalase activities by 56%, 44%, and 53% in shoots and 62%, 49%, and 65% in roots, respectively, indicating a robust defense mechanism against Cd stress. Notably, OsNramp5, OsHMA3, OsSOD-Cu/Zn, OsCATA, OsCATB, and OsAPX1 showed significant expression after SiO NPs treatment, suggesting potential Cd translocation within rice tissues. Overall, SiO NPs supplementation holds promise for enhancing Cd tolerance in rice plants while maintaining essential physiological functions.

Safe levels of heavy metals in the surface water and sediment of the eastern Bay of Bengal coast have not been universally established. Current study characterized heavy metals such as arsenic (As), chromium (Cr), cadmium (Cd) and lead (Pb) in surface water and sediments of the most important fishing resource at the eastern Bay of Bengal coast, Bangladesh. Both water and sediment samples were analyzed using inductively coupled plasma mass spectrometer. Considering both of the seasons, the mean concentrations of Cr, As, Cd, and Pb in water samples were 33.25, 8.14, 0.48, and 21.14 µg/L, respectively and in sediment were 30.47, 4.48, 0.20, and 19.98 mg/kg, respectively. Heavy metals concentration in water samples surpassed the acceptable limits of usable water quality, indicating that water from this water resource is not safe for drinking, cooking, bathing, and any other uses. Enrichment factors also directed minor enrichment of heavy metals in sediment of the coast. Other indexes for ecological risk assessment such as pollution load index (PLI), contamination factor (CF), geoaccumulation index (Igeo), modified contamination degree (mCd), and potential ecological risk index (PERI) also indicated that sediment of the coastal watershed was low contamination. In-depth inventorying of heavy metals in both water and sediment of the study area are required to determine ecosystem health for holistic risk assessment and management.

Meretrix lyrata which is under the family of Veneridae and under the order of Venerida, is a nutritionally and economically important edible mussel in Bangladesh. However, studies on species identification and nutritional value in M. lyrata are scarce. Therefore, a detailed investigation was conducted on (i) species identification of the common edible mussel through DNA-barcoding and morphometrics, (ii) reproductive features, such as size at sexual maturity, spawning, and peak-spawning seasons under different environmental factors, and (iii) nutritional status through proximate analysis of M. lyrata mussel collected from the Bay of Bengal, Bangladesh. The results indicated that the size at sexual maturity for M. lyrata was 4.2 cm and the spawning seasons were significantly affected by the dissolve oxygen and salinity. The study also demonstrated that the spawning of M. lyrata occurred from January to June and December while peak spawning season was May in the Bay of Bengal. The higher protein and moisture contents with lower fat in M. lyrata indicated that are value-added seafood with higher nutritional values for consumers.

Coastal vegetative ecosystems are among the most threatened in the world, facing multiple anthropogenic stressors. A good example of this is seagrass, which supports carbon capture, coastal stabilization, and biodiversity, but is declining globally at an alarming rate. To understand the causes and consequences of changes to these ecosystems, we need to determine the linkages between different biotic and abiotic components. We used data on the seagrass, Zostera marina, collected by citizen scientists across 300 km of the south coast of the United Kingdom as a case study. We assembled data on seagrass genotype, phenotype, infauna, and associated bathymetry, light, sea surface temperature, and wave and current energy to test hypotheses on the distribution and diversity of this temperate sub-tidal ecosystem. We found spatial structure in population genetics, evident through local assortment of genotypes and isolation by distance across a broader geographic scale. By integrating our molecular data with information on seagrass phenotype and infauna, we demonstrate that these ecosystem components are primarily linked indirectly through the effects of shared environmental factors. It is unusual to examine genotypic, phenotypic, and environmental data in a single study, but this approach can inform both conservation and restoration of seagrass, as well as giving new insights into a widespread and important ecosystem.

The Plicosepalus genus includes hemiparasitic mistletoe and belongs to the Loranthaceae family, and it has several medicinal uses. In the present study, we sequenced the complete plastomes of two species, Plicosepalus acaciae and Plicosepalus curviflorus, and compared them with the plastomes of photosynthetic species (hemiparasites) and nonphotosynthetic species (holoparasites) in the order Santalales. The complete chloroplast genomes of P. acaciae and P. curviflorus are circular molecules with lengths of 120,181 bp and 121,086 bp, respectively, containing 106 and 108 genes and 63 protein-coding genes, including 25 tRNA and 4 rRNA genes for each species. We observed a reduction in the genome size of P. acaciae and P. curviflorus and the loss of certain genes, although this reduction was less than that in the hemiparasite and holoparasitic cp genomes of the Santalales order. Phylogenetic analysis supported the taxonomic state of P. acaciae and P. curviflorus as members of the family Loranthaceae and tribe Lorantheae; however, the taxonomic status of certain tribes of Loranthaceae must be reconsidered and the species that belong to it must be verified. Furthermore, available chloroplast genome data of parasitic plants could help to strengthen efforts in weed management and encourage biotechnology research to improve host resistance.

Wildfires significantly alter soil properties and result in vegetation shifts; therefore, rapid reforestation activities are needed in the forests affected by wildfires. The decreased nutrient in the soil is the obvious effect of wildfires; however, little is known about the reforestation of Juniper (Juniperus procera) forests with application of NPK fertilizers. Juniper forests are common in Asir and Taif regions of Saudi Arabia and vulnerable to wildfires; thus, reforestation is needed after the onset of fires. This study assessed the impact of different doses of organic NPK fertilizer (0, 5 and 10 g/L) on growth and nutrient accumulation of Juniper trees grown on fire-damaged and intact soils. Data relating to tree height, number of leaves per plant, fresh and dry biomass accumulation in shoot and root, chlorophyll contents and uptake of N, P, K, and Na were recorded. Individual and interactive effects of soil types and fertilizer doses significantly altered all measured traits with minor exceptions. Overall, higher values of the measured traits were recorded for intact soil and 10 g/L fertilize dose. The increasing fertilizer doses improved the growth and nutrient acquisition and application of 10 g/L fertilizer on intact soil recorded the highest values of growth traits. Juniper trees grown on fire-damaged soil accumulated higher amount of nitrogen than intact soil. Similarly, the trees grown on intact soil accumulated lower amount of Na and maintained comparable K/Na ratio to intact soil. It is concluded that supplying 10 g/L fertilizer could improve the establishment of Juniper trees on fire-damaged soil. Therefore, organic fertilizer can be used to improve the reforestation of wildfire-affected Juniper forests in the Asir province.

Environmental contamination caused by various pollutants due to automobile emissions is an alarming issue. One important type of the pollutants are heavy metals, including chromium (Cr) added by the exhaust of toxic smoke of vehicles. These pollutants are added to forage crops cultivated near roadsides, soil and irrigation water. However, rare studies have been conducted to infer Cr accumulation near heavy automobile emission areas. This study was conducted to determine Cr concentration in irrigation water, soil and forage. Water, forage and soil samples were collected from area impacted by heavy traffic. Atomic absorption spectrophotometer was used to appraise Cr values in the collected samples. Chromium values ranged from 0.50 to 1.14 mg/kg in water samples and from 0.04 to 2.23 mg/kg in soil samples. It was highest in Zea mays grown soil, whereas minimum in Brassica campestris soil. The Cr values in forages ranged from 0.09 to 1.06 mg/kg. Z. mays observed the highest Cr accumulation, whereas the lowest Cr accrual was noted for B. campestris. The pollution load index (PLI) was the highest for Trifolium alexandrinum, while the lowest for Z. mays. Bio-concentration factor (BCF) ranged from 0.14 to 8.63. The highest BCF was noted for T. alexandrinum, while the lowest for Z. mays. The highest and the lowest daily intake of metal (DIM) was noted for Z. mays at different sites. Health risk index (HRI) was highest for Z. mays and lowest for B. campestris. The results add valuable information on heavy metal accumulation in water, soil and forage samples near to automobile emission area.

Individual effects of application of nitrogen (N) and plant densities (PD) were reported in various studies; however an interactive effect of N and PD in cotton was not studied. To explore the benefits of interactive effects of N fertilizer and PD to increase the quality of cotton. This study was carried out in randomized complete block design (RCBD) with split plot arrangement. In split plot arrangement, main plot was consisted of N application rate and in sub plots different PD were done. There were two nitrogen levels; low N level (F1) 120 kg ha-1 and high N level (F2) 180 kg ha-1 and three planting densities; 8 plants m-2 as low density (LD), 10 plants m-2 as medium density (MD) and 12 plants m-2 as high density (HD). In this study we observed the interactive effect of N application levels and PD on cotton photosynthetic and agronomic traits of various stages of development. Results showed that cotton growth and N contents was varied among treatments on different development stages. Plant biomass production, photosynthetic rate (Pn), intercellular CO2 (Ci), water use efficiency (WUE) and N contents were unaffected at the seedling stage by N application rate and PD, however, the highest Pn, Ci and N contents was at squaring stage followed by blooming stage. Higher seed cotton yield and lint yield were obtained F1 with HD, and F2 with MD yielded the highest N contents and cotton yield among treatments. We found that the squaring stage was more critical, followed by the blooming stage when considering N rate and PD.

Soil salinity adversely affects the growth, yield, and quality parameters of sugar beet, leading to a reduction in root and sugar yields. Improving the physical and chemical properties of salt-affected soils is essential for sustainable cultivation and sugar beet production. A field experiment was conducted at the Delta Sugar Company Research Farm, El-Hamool, Kafr El-Sheikh, Egypt, to evaluate the response of sugar beet to the application of beet sugar filter cake treated with sulfuric and phosphoric acid-treated, phosphogypsum (PG), desaline, humic acid, and molasses under saline soil conditions. The application of treated filter cake enhanced root length, diameter, and leaf area. The application of molasses enhanced root length, diameter, and leaf area as well. Application of molasses increased sugar content and root yield. The application of either treated filter cake or molasses produced the highest recoverable sugar yield. Linear regression analysis revealed that the root yield, quality index, and recoverable sugar yield increased in response to the increased availability of either Ca2+ or K content in the soil which increases in response to the application of soil amendments and molasses. The application of treated beet sugar filter cake and molasses increased the calcium, magnesium, and potassium availability in the soil. Treated filter cake is a promising organic soil amendment that enhanced the yield by 29% and yield-related traits of sugar beet by improving the physical and chemical properties of the soil.

Salinity is among the major abiotic stresses negatively affecting the growth and productivity of crop plants. Sodium nitroprusside (SNP) -an external nitric oxide (NO) donor- has been found effective to impart salinity tolerance to plants. Soybean (Glycine max L.) is widely cultivated around the world; however, salinity stress hampers its growth and productivity. Therefore, the current study evaluated the role of SNP in improving morphological, physiological and biochemical attributes of soybean under salinity stress. Data relating to biomass, chlorophyll and malondialdehyde (MDA) contents, activities of various antioxidant enzymes, ion content and ultrastructural analysis were collected. The SNP application ameliorated the negative effects of salinity stress to significant extent by regulating antioxidant mechanism. Root and shoot length, fresh and dry weight, chlorophyll contents, activities of various antioxidant enzymes, i.e., catalase (CAT), superoxide dismutase (SOD), peroxidase (POD) and ascorbate peroxidase (APX) were improved with SNP application under salinity stress compared to control treatment. Similarly, plants treated with SNP observed less damage to cell organelles of roots and leaves under salinity stress. The results revealed pivotal functions of SNP in salinity tolerance of soybean, including cell wall repair, sequestration of sodium ion in the vacuole and maintenance of normal chloroplasts with no swelling of thylakoids. Minor distortions of cell membrane and large number of starch grains indicates an increase in the photosynthetic activity. Therefore, SNP can be used as a regulator to improve the salinity tolerance of soybean in salt affected soils.

This study provides a taxonomic revision for Ceropegia sect. Huernia in the flora of Saudi Arabia. Forty-six quantitative and qualitative morphological characters were analysed using principal component analysis (PCA), principal coordinates analysis (PCoA) and the unweighted pairs group using mean average (UPGMA) to separate and help delimit taxa. We propose to reduce the number of species reported in Saudi Arabia from 11 to four: C. khalidbinsultanii comb. nov., C. laevis, C. lodarensis and C. macrocarpa. This study also suggested reducing two names to varietal level under C. lodarensis (var. foetidacomb. nov. and var. rubrostictacomb. nov.). A key to the species, detailed morphological descriptions, illustrations, distribution maps, ecology, etymology and preliminary conservation assessments are provided that follow IUCN criteria.

An endangered and rare species Aloe pseudorubroviolacea from the plant family Asphodelaceae which is presently recorded as endangered in Saudi Arabia collected from Al-Baha region of Saudi Arabia its GPS Latitude and Longitude coordinates 19.8345, 41.5481. The chloroplast matK and rbcL gene was considered in this study based on molecular identification the size is about 571 and 664 bp respectively. From the sequence analysis the gene matK and rbcL confirm that this species is very much closely related with A. rubroviolacea and also inter related with the species Astroloba rubriflora, Chrysopogon gryllus, Chortolirion angolense shows about 98.7% sequence homology. The partial matK and rbcL gene sequence discriminate Aloe pseudorubroviolacea from the closely related plant species, A. rubroviolacea. The gene sequence of rbcL discriminates the species from Chrysopogon gryllus and Chortolirion angolense, demonstrates the nucleotide variations in 3 different sites (623C/T; 653C/T; 700C/A). This study showed that matK and rbcL sequence region of chloroplast gene used to authenticate the samples of A. pseudorubroviolacea and which provide to help in correct identification and conservation process of this medicinally valuable endangered plant species.

RESUMO

Strongyloidiasis is an infection caused by the intestinal nematode Strongyloides stercoralis. Infected healthy individuals are usually asymptomatic, however it is potentially fatal in immunocompromised hosts due to its capacity to cause an overwhelming hyperinfection. Strongyloidiasis could be missed during routine screening because of low and intermittent larval output in stool and variable manifestations of the symptoms. We present two cases of strongyloidiasis occurring in children with solid organ malignancies suspected to have the infection based on their clinical conditions and treatment history for cancer. Both patients were diagnosed by molecular and serological tests and were successfully treated. Thus, strongyloidiasis in patients undergoing intensive treatment for malignancies should be suspected, properly investigated and treated accordingly.

In this study we have cloned unreported gene fragments of Toxoplasma gondii GRA7 and SAG1 and expressed the corresponding recombinant proteins, followed by evaluation of their usefulness for the serological diagnosis of toxoplasmosis. Both recombinant proteins were expressed efficiently in insoluble form, purified by single step Ni-NTA affinity chromatography and their antigenicity to detect toxoplasma specific IgG antibodies were determined by immunoblotting. A total of 60 serum samples from three groups of individuals based on their anti-toxoplasma antibody profiles were tested, namely (I) IgM+, IgG+ (n=20), (II) IgM-, IgG+ (n=20) and (III) IgM-, IgG- (n=20). Both recombinant proteins exhibited high sensitivity (100%) with sera from Group I. rGRA7 and rSAG1 reacted 40% and 80% respectively with Group II sera. The specificity of the recombinant proteins based on reactivities with Group III sera were 100% and 80% with rGRA7 and rSAG1 respectively. Thus rGRA7 was found to be better at discriminating probable acute from chronic phases of toxoplasmosis, and it also showed higher specificity.

INTRODUCTION: Strongyloides stercoralis infection can persist in the host for several decades, and patients with cancer and other clinical conditions who are exposed to immunosuppressive therapy are at risk of developing hyperinfection. CASE REPORT: This is a case of angioimmunoblastic T-cell lymphoma (AITL) in a patient with lymphadenopathy and bulky neck mass. Severe sepsis and episodes of diarrhea were observed upon the first cycle of cyclophosphamide, doxorubicin, oncovin (vincristine) and prednisone (CHOP) regime chemotherapy preceded by high dose of dexamethasone. There was Klebsiella pneumoniae bacteremia and moderate eosinophilia. Rhabditiform S. stercoralis larvae were observed in the stool, and this was confirmed by real-time PCR. Strongyloides-specific IgG and IgG4 were also positive. The patient was treated with oral albendazole (400mg/day) for 3 days and intravenous tazocin (4.5gm/6 hours) for 5 days; however he succumbed following multi-organ failure. CONCLUSION: This is likely a case of Strongyloides hyperinfection with secondary bacteremia.
